WT (XL2001) and the hfq R16A mutant (FDM1050) were transformed with either an empty vector (pBAD24) or a T4 RNA Ligase I-expressing vector (pBAD24- t4rnl1 ). Biological duplicates of each strain were cultured in LB (Lennox) with ampicillin at 37°C until reaching an OD600 of ∼0.5, at which point arabinose was added to induce enzyme expression.
